Immunohistochemical localization of two alpha- and one beta-glycoprotein subunits of the human alpha1-antitrypsin-protease inhibitor complex in nonneoplastic tissues. In this study, the expression of the two alpha- and one beta-glycoprotein subunits of the alpha1-antitrypsin-protease inhibitor complex was immunohistochemically evaluated in nonneoplastic human tissues. In addition, expression of cytochrome P450 3A4 was also evaluated. Antisera were raised against the alpha1-antitrypsin and the beta-glycoprotein. For evaluation of protease inhibitor expression, the two subunits were applied as single proteins and as a complex. Concerning the expression of the alpha1-antitrypsin-protease inhibitor complex the following results were obtained: The hepatocytes and the transitional epithelium of the kidney, the epithelium of the intestine, the gallbladder, the bronchioles, the exocrine pancreas, the basal cells of the stomach, and the thymus were strongly positive for the alpha1-antitrypsin-protease inhibitor complex, whereas the smooth muscle of the gut and the arterial vessel wall were only weakly positive. For the beta-glycoprotein, the epithelium of the intestine and the mucosa of the stomach was strongly positive, whereas the smooth muscle of the gut was weakly positive. For the evaluation of cytochrome P450 3A4, only a weak staining was detected in hepatocytes of the liver. The data obtained indicate that the hepatocytes and the transitional epithelium of the kidney, the epithelium of the intestine, the gallbladder, the bronchioles, the exocrine pancreas, the basal cells of the stomach, and the thymus are most likely to be involved in the synthesis of the alpha1-antitrypsin-protease inhibitor complex in humans.
